The holy grail myth comes from Arthurian legends, the origins of which are not fully known.

The Ark of the Covenant is an entirely different legend from the bible, book of Exodus. The story goes, when the Philistines captured it, they suffered terrible sickness, death and destruction. That God would destroy any evil people who came close to it.

There's a great alternate, ancient aliens, theory that the ark actually contained a radioactive energy generator, that only trained handlers (Israelites trained by aliens) could safely handle and use. The aliens gave to them to help them through their 40 years in the desert.

The Philistines, not having the training to handle the device, unleashed a huge amount of radiation which killed anyone in immediate proximity and gave radiation sickness to the surrounding population. The Ark myth has inspired some fun theories:
